Résumé In order to develop a complete understanding of a biological system, information must cover multiple dimensions. Over the last ten years, we have witnessed decisive advances in bioinformatics, genome sequencing, and high-throughput technologies, that have highlighted the need for approaching biological systems as a whole. Metabolic diseases, including type 2 diabetes and cardiovascular disease, as well as cancer, involve complex genetic, molecular, and environmental interactions, and systems-based approaches have proven to be instrumental in tackling this complexity by integrating genomic, molecular, and physiological data. This meeting will provide a unique opportunity to bring together experts in systems biology and metabolism to discuss how ‘Omics’ approaches can be exploited in an effort to understand the perturbations that take place in the pathogenesis of metabolic diseases. We will discuss novel approaches for studying metabolic alterations in a high-throughput scale and explore how epigenomics, non-coding RNAs, and environmental factors control metabolic pathways in disease settings.

Résumé Throughout most of the latter half of the 20th century, research on RNAs focused predominantly on their key role as intermediaries between DNA and protein. It was Jacob and Monod who originally proposed that RNAs, because of their intrinsic specificity, might be the elusive factors that instruct genes to turn on and off. In the end, these regulatory factors turned out to be proteins, now commonly referred to as transcription factors. Yet, with the discovery of small RNAs in the 1990s, a wave of new research on RNAs has resurrected the idea that RNAs can also serve regulatory functions.

Résumé Nucleic acids – DNA and RNA – encode and transmit genetic information in the cell. But they are also structural, regulatory, and can even behave like enzymes. DNA is a repository of genetic information and must be accurately copied once and only once in each cell cycle. Structurally it has to be packaged into chromosomes yet accessible when required. DNA is subjected to recombination that creates diversity and leads to evolution, while it must remain intact and undergoes repair when damaged. RNA by contrast is the ‘working substance’ of genetics and a truly versatile molecule. It is a messenger that goes between DNA and protein, the scaffold for building complex functional machines such as the ribosome, a molecular switch responding to small molecules, and a key player in complex regulatory events. RNA can even accelerate chemical reactions by a million fold or more. Understanding processes involving DNA and RNA at the molecular and chemical level is the major theme of this conference.

Résumé This meeting brings together a broad range of researchers developing and using optogenetic probes with researchers developing related technologies. Specific goals include to: 1) Present the latest innovative new genetically-encoded tools and technologies for manipulating biological systems with light; 2) Promote exchange between those developing light-responsive probes and developers of related tools (e.g., biosensors, hardware) that will maximize the power of optogenetics; and 3) Foster the exchange of ideas between optogenetic tool developers and users.

Résumé Elastin, Elastic Fibers and Microfibrils conference is now recognized as the major international meeting for investigators with an interest in the biochemistry, cellular and molecular biology, and pathology of elastin. Over the years to follow, the breadth of the field has grown even more due to identification and study of numerous additional molecules that influence the regulation, assembly and function of elastic fibers, such as the fibulins, latent TGF-beta binding proteins, emilins, proteoglycans and lysyl oxidases. The Conference is unique in the depth of its coverage of elastin biology, spanning from detailed, high-resolution structural studies of elastin and fibrillin, to in vitro assembly assays, to functional in vivo research with transgenic and knockout animals, to human disease, and finally to biomaterials and therapies. The unusual biology of elastic fibers has brought together basic scientists with diverse areas of expertise, including physical chemistry, engineering, cell and molecular biology, developmental biology, biochemistry and biotechnology. Furthermore, because of the involvement of elastin and elastic fiber associated proteins in numerous pathological processes, past Gordon Conferences have attracted investigators with a wide spectrum of clinical interests, including emphysema, COPD, pulmonary and vascular hypertension, atherosclerosis, aneurysms, genetic diseases, and cutaneous and eye diseases. Résumé The Cell Biology of Metals Gordon Conference was initiated to provide a forum for an emerging field, encompassing the uptake, distribution and use of metals in cells and organelles. Metals are taken up across membranes and are directed to various cellular destinations, ultimately finding their way into the correct metalloproteins. Intricate mechanisms control uptake, distribution, trafficking, and insertion into proteins. Transporters, chelators, metal cofactors, protein folding chaperones, metallochaperones, transcription factors and translation regulators are involved. The level of complexity equals that of the protein trafficking pathways that are being characterized by modern methods of cell biology. An important objective is to understand how protein trafficking pathways and metal trafficking pathways are integrated and intersect. Other areas of focus are mechanisms of metal ion homeostasis, how cells sense diverse metal ions, and how homeostatic mechanisms for different metal ions are interconnected. Cellular malfunction or disease ensue if any of the steps in metal homeostasis are perturbed, and the meeting emphasizes these disease related implications. The sessions are organized according to cell structure, cutting across metals (Fe, Cu, Zn, Mo) and evolutionary categories.

Résumé The Gordon Research Conference on Cellular and Molecular Mechanisms of Toxicity is a premier forum for showcasing the latest, most innovative advances in mechanistic toxicological research. For the 2015 conference, the organizers have assembled a group of world-leading experts working in areas of investigation that are highly relevant to environmental, industrial, and pharmaceutical toxicology, with the goal of incorporating research on the mechanistic basis of disease into risk decision making. Emerging technologies in mechanistic toxicology and computational, 3D, and in vivo modeling as well as systems biology approaches to assessing risk to human disease will be highlighted. The microbiome, the inflammasome, epigenetics, carcinogenesis, and stem cells in toxicological research are among the topics that will be highlighted at the conference. The conference will attract a mix of junior and senior investigators and trainees with a common interest in advancing their respective fields of investigation. For the second time, a Gordon Research Seminar on Cellular and Molecular Mechanisms of Toxicity will precede the conference. This provides an expanded opportunity for trainees and young scientists to be exposed to cutting-edge science and to network with their future colleagues in the field. The poster sessions are an essential element to the success of conference. Trainees and scientists at any level will have the opportunity to present their latest, groundbreaking work in poster format under an informal atmosphere that fosters relaxed dialog, networking and idea sharing. A special committee will select some abstracts/posters for short oral talks. These "late-breaking" oral sessions provide increased participation to trainees and young scientists to share their work with an audience of experts. This is a tremendous opportunity for the young talent to receive feedback from well-established leaders in the field. Another key to the success of the Gordon Conference is the social interaction of participants at meal times and during social activities in afternoon breaks. These interactions have led to long-lasting scientific collaborations and the lining up of trainees with laboratories for further education.
