Over the last ten years, we have witnessed decisive advances in bioinformatics, genome sequencing, and high-throughput technologies, that have highlighted the need for approaching biological systems as a whole. Metabolic diseases, including type 2 diabetes and cardiovascular disease, as well as cancer, involve complex genetic, molecular, and environmental interactions, and systems-based approaches have proven to be instrumental in tackling this complexity by integrating genomic, molecular, and physiological data. This meeting will provide a unique opportunity to bring together experts in systems biology and metabolism to discuss how ‘Omics’ approaches can be exploited in an effort to understand the perturbations that take place in the pathogenesis of metabolic diseases. We will discuss novel approaches for studying metabolic alterations in a high-throughput scale and explore how epigenomics, non-coding RNAs, and environmental factors control metabolic pathways in disease settings. In the end, these regulatory factors turned out to be proteins, now commonly referred to as transcription factors. But they are also structural, regulatory, and can even behave like enzymes. DNA is a repository of genetic information and must be accurately copied once and only once in each cell cycle. Structurally it has to be packaged into chromosomes yet accessible when required. DNA is subjected to recombination that creates diversity and leads to evolution, while it must remain intact and undergoes repair when damaged. RNA by contrast is the ‘working substance’ of genetics and a truly versatile molecule. It is a messenger that goes between DNA and protein, the scaffold for building complex functional machines such as the ribosome, a molecular switch responding to small molecules, and a key player in complex regulatory events. RNA can even accelerate chemical reactions by a million fold or more. Understanding processes involving DNA and RNA at the molecular and chemical level is the major theme of this conference. Metals are taken up across membranes and are directed to various cellular destinations, ultimately finding their way into the correct metalloproteins. Intricate mechanisms control uptake, distribution, trafficking, and insertion into proteins. Transporters, chelators, metal cofactors, protein folding chaperones, metallochaperones, transcription factors and translation regulators are involved. The level of complexity equals that of the protein trafficking pathways that are being characterized by modern methods of cell biology. An important objective is to understand how protein trafficking pathways and metal trafficking pathways are integrated and intersect.
