Keystone Symposia: Precision Genome Engineering and Synthetic Biology

Résumé The most exciting recent advances in viral immunity include the increased understanding of the role of innate immune mechanisms, interaction between innate and adaptive immunity, pathogen manipulation of host responses, activation of immune responses and immune effector functions. Despite these advances, basic mechanisms of anti-viral immunity are poorly characterized for many acute infections and less well understood for chronic infections. This meeting will include both innate and adaptive immunity and juxtapose animal models with human studies as it attempts to foster collaborative efforts between attending viral immunologists working in disparate systems and models.

Résumé The objective of the 2015 Directed Cell Migration Gordon Conference is to bring together an international group of young and senior scientists that share a common interest in cell migration in an atmosphere designed to foster new interdisciplinary approaches and excitement for young scientists. This conference will provide an environment to develop interactions among scientists from multiple disciplines that are related to directed cell migration, including participants from the fields of immunology and inflammation, cancer invasion and metastasis, embryological development and tissue repair, as well as related fields, including engineering, physics and computational biology. This high level of scientific interaction will encourage the attendees to learn about latest research findings, develop novel hypotheses, and enhance insight into how cell migration contributes to disease. The session speakers, selected from a variety of fields, will emphasize novel, unpublished results directly related to gradient sensing and directed cell migration. Young investigators will be encouraged to present and discuss their findings in a supportive environment through the invitation of up to 20 additional speakers, selected from submitted abstracts, and by participation in the posters sessions. Throughout the conference, interactions between senior and young investigators will be promoted in both formal and informal settings. To maximize interactions and discussions, each session of the 2015 Conference is designed to have a mixture of talks from biologists and physicists/engineers/mathematicians. This will enhance the collegiality of the conference and help build collaborations between different fields.

61.Gordon Research Conference — Cilia, Mucus and Mucociliary Interactions 2015

Résumé Since its inception, the GRC on Cilia, Mucus, and Mucociliary Interactions has provided a unique forum that brings together researchers from multiple closely related disciplines. Topics revolve around the latest information on basic mechanisms of cellular assembly, processing, and function, and the relationship between motile cilia, mucus and related features that are an essential component of fundamental biological processes. As such, issues related to multiple disciplines are embraced. The result is a highly integrated, international gathering of basic scientists, bioengineers, translational investigators, and clinicians who study the components of the mucociliary apparatus, the fundamentals of cilia and mucus biology, and their interface. We will continue the tradition of featuring cutting-edge research of motile cilia and mucus as related to diseases including cystic fibrosis, primary ciliary dyskinesia, chronic rhinosinusitis, asthma, and COPD, as well as heterotaxy and congenital heart disease. The 2015 meeting will address specific gaps in our knowledge such as: - What are the cellular and molecular origins of newly identified genetic and acquired dysfunction of cilia, mucin and mucus? - How can we image components, functions and interactions of cilia and mucus at the molecular level with high resolution? - How do defects in mucus and cilia impact mucosal immunity? - As we translate basic discoveries, what diagnostic and therapeutic approaches can monitor and correct defects? The meeting will open with keynote talks from eminent scientists who will detail the current state-of-the-art and paradigms, and articulate the key challenges for the future, while subsequent sessions will focus on specific issues in the field. For each session, discussion leaders will provide a brief introduction and perspective on the topics to be covered. Established and up-and- coming scientists will present new unpublished data, as is the tradition of GRCs. Sessions will include speakers from inside and outside the field. We also include an evening of "late breaking science" composed of six talks selected from submitted abstracts. In addition to the platform presentations, afternoon poster sessions with "poster rounds" will highlight exceptional work and enhance investigator-trainee discussions. Abstracts from young investigators and trainees selected for short talks or with highlighted posters will be awarded travel grants.

66.EMBO Practical Course — In vivo plant imaging

Résumé This course will teach participants the practical aspects of live-imaging plant tissues. Plants are quite different to animals in many ways. For instance they typically develop much more slowly and cells do not migrate. Their overall morphologies are also very different and the presence of cell walls presents challenges optically. Over many years, certain laboratories have developed specialised imaging techniques optimised for plants. However despite these advances, the adoption of these techniques by the wider plant research community has been extremely limited. These specialised techniques will be taught to help promote their wider adoption. The course will cover the physical preparation of samples from different parts of the plant as well as time-lapse imaging strategies and image analysis approaches and uses.

Résumé Fluorescence is a light-based molecular phenomenon that is inherently non-invasive; it thus finds broad application in the studies of living matter - from the observation of isolated proteins in buffer to complex machinery in living animals. Detection of fluorescence offers highest signal to noise, such that even single molecules can be investigated. This coined the development of myriads of fluorescence-based experimental strategies in biology and biophysics. Progress in biological research is directly linked to the development and subsequent application of new spectroscopic/ microscopic techniques. Researchers in biology are mostly question driven – their aiming at using novel light-microscopy often reflects the hope to gain new insights, based on the promises of publication on the methods. This typically causes a misperception, e.g. that certain experiments performed under controlled biochemical conditions might not be easily adaptable to studies in cells, or that high-resolution methods do frequently not match the demands of in cell studies. Vice versa, in cell studies frequently lack the insights into the molecular mechanism. To achieve the necessary understanding to integrate seamlessly fluorescence tools across different resolution levels and laboratory scales (biochemical-cell-organism) in their research projects, the participants will receive hands on training and specialized training across a large range of high-resolution fluorescence studies. In particular, the course aims to teach the common concepts but also what requirements distinguish a large set of advanced fluorescent spectroscopic and microscopic techniques, from biochemical conditions to complex cellular systems. Experts in different fields will introduce principles, challenges and guidance of their fields. In this surrounding the participants will be able to directly perform small research projects in teams of two people on state of the art equipment and discuss their results with invited investigators, instructors, local teachers and editor.

Résumé An ambitious goal in modern biology is to understand stem cell states. Although much is known about both the behavior of stem cells under various conditions and the molecular details of stem cell identity, there is a gap between these two bodies of knowledge. This meeting sets out to bridge this gap, with a focus on transcriptional and epigenetic control of stem cell function. Advances in these areas will enable us to devise new strategies not only to probe stem cell biology more deeply, but to model disease and to develop cellular therapies. This Keystone Symposia meeting will bring together researchers who strive to: 1) Describe transcriptional and epigenetic mechanisms that control specific stem cell behaviors; 2) Generate predictive models of stem cell fate determination; 3) Delineate the influence of cell cycle and signaling on stem cell state; 4) Understand mechanisms governing cell fate change, including differentiation, reprogramming and transdifferentiation. Ultimately, the meeting will provide an opportunity to view copious empirical data on stem cell behavior through a molecular lens. Résumé The causative links between genomic and genetic variations and cancer are well documented and have provided a greater understanding of tumorigenesis and cancer progression. Molecular genetic information has been used to derive new therapeutic regimens and has accelerated the development and application of "personalised cancer medicine". There are also an increasing appreciation that epigenetic aberrations that are often directly caused by genetic defects resulting in loss- or gain-of-function of epigenetic-regulators also contribute significantly to cancer onset and progression. Indeed, parallel integration of our knowledge of the cancer genome and epigenome using sophisticated "omics-based" technologies and high throughput functional screening techniques are providing even more tangible links between genetic/genomic aberrations, epigenetic/epigenomic dysregulation and tumorigenesis. This information is being functionally validated using experimental models that accurately reflect the underlying genetic and epigenetic alterations observed in human tumor samples. Moreover, the information is being translated into clinical use through the development of genetic and epigenetic biomarkers of cancer development, evolution, heterogeneity and response to therapeutic intervention. This GRC series focuses on the advances in discovering and validating "driver" mutations in cancer genes, understanding the downstream consequences of these genetic changes on the epigenome and how this information is being best utilized to advance cancer detection, monitoring and therapy.

Résumé What is the relationship between normal tissue stem cells and tumor-initiating cells (e.g., cancer stem cells)? Which signaling and other regulatory networks are altered in tumors relative to the normal tissues, and how do they function within the tumor? Finally, there is growing evidence that therapies aimed at the major cell types in tumors may sometimes make things worse, by leading to an expansion in the fraction of cancer stem cells. How can this be avoided? This workshop will address these and other questions through discussions among mathematical and computational modelers and experimentalists. In particular, the strong connections between normal development, tumor growth and the use of novel treatment strategies will be discussed.

Fusing new developments in basic science with high-impact translational research, the meeting will provide a stimulating program featuring an international mix of established and upcoming researchers. The meeting will start with two Keynote speakers, Drs. John Hartwig and Shaun Jackson, covering ground-breaking aspects of megakaryocyte and platelet biology. Meeting topics will include emerging roles for platelets in biology, including immunity, tumor metastasis, and lymphatic development. Identification of new signaling pathways that regulate megakaryocyte development and platelet activation will also be a key theme, including an original session focused on membrane biology. Novel mechanisms for growing platelets in vitro as well as innovative ways to visualize and study megakaryocytes and platelets will also come under the spotlight. The conference will also report on the latest findings in the regulation of platelet numbers. In addition, there is an entire session devoted to presentations of Late-Breaking Topics, selected from submitted abstracts. Building on the highly creative, congenial interactions that characterized the previous meetings in this series, this conference will provide the ideal setting for cross-fertilization of ideas, conceptual advances, and the promotion of synergistic collaborations.

Résumé The 2015 Gordon Research Conference (GRC) on "Cannabinoid Functions in the CNS: From Molecules to Disease Mechanisms" will celebrate the 50th anniversary of the discovery of the chemical structure of Δ9-tetrahydrocannabinol, the major psychoactive constituent of Cannabis sativa. This conference will also mark the 25th anniversary of cloning the "neuronal" CB1 cannabinoid receptor. Thus, the meeting will blend the review of long-held conceptual milestones that provided molecular fundaments for endocannabinoid research performed today with disseminating and discussing the newest, groundbreaking results among students, junior faculty and senior investigators. Besides capturing physiological roles for endocannabinoids, ranging from secondary metabolism, synapse physiology to developmental biology including stem cells, this GRC will also serve as a platform for presenting key insights in fear, anxiety, psychosis, addiction, reward, aging and neurodegeneration. Our stated aim is to emphasize molecular mechanisms and comprehensive studies establishing causal relationships to endocannabinoid signaling. Significantly, this topical GRC will feature introductory lectures from world-renown investigators outside the immediate endocannabinoid field to promote interdisciplinarity, interactions, and the discussion of theoretical and technological advances pertinent to advancing endocannabinoid neurobiology. As such, the opening lecture will feature Tamas L. Horvath, while plenaries at the various sessions include Elena Cattaneo, Karl Deisseroth, Andreas Luthi, Raphael Mechoulam and Pier Vincenzo Piazza. We actively seek the engagement of prospective leaders to this exciting research area. Poster presentations will be held on all conference days, providing an outstanding opportunity for early career investigators to discuss their results in an expert environment. All prospective speakers have committed to emphasize unpublished results.

Résumé About 50-200 billion of our cells die daily by apoptosis or programmed cell death in the human body. The mechanisms of apoptosis have been extensively studied, but those of the elimination of apoptotic cells are much less understood. Apoptotic cells are rapidly engulfed and digested by their neighbors or by professional phagocytes in a process known as phagocytosis, clearance or efferocytosis. During development this process plays a key role in tissue morphogenesis and renewal. It also allows for the maintenance of tissue homeostasis in adults and promotes the resolution of inflammation. Failure to clear apoptotic cells can result in developmental defects, chronic inflammatory disorders, autoimmune and neurodegenerative diseases. For example, it has been associated with Lupus, a chronic inflammatory autoimmune disease, and with retinal degeneration. Dying cells can be cleared before completion of their programmed death, and failure in this process may result in cancer. A thorough understanding of efferocytosis is essential to design new therapies to prevent or treat chronic inflammation, autoimmune and neurodegenerative diseases, and cancer. Some of the molecular mechanisms of apoptotic cell clearance are shared with those of pathogen clearance, and thus studying efferocytosis will also lead to new therapies to fight infections. This is critical since pathogens are becoming resistant to current treatments. At this Gordon Research Conference on Apoptotic Cell Recognition & Clearance, we will discuss the latest advances made in our understanding of the molecular mechanisms of apoptotic cell recognition and clearance in all model organisms, from worm, fly, fish and mouse, to mammalian (including human) cells in the context of both healthy and disease states.

Sessions will focus on basic events underlying cell division. Résumé From genomes to biomes, from microbes to plants and animals, the 2015 Gordon Research Conference on Ecological and Evolutionary Genomics will highlight how genome-enabled approaches are helping to rapidly advance our understanding of the complicated relationship between genotype, phenotype and the environment. Topic areas such as genome evolution, microbiomes & symbiosis, speciation & adaptation, phylogenomics and organismal responses to climate change, will highlight how biodiversity can be used to illuminate complex biological relationships and inform ecological and evolutionary processes and molecular mechanisms of adaption to changing environments. The conference will also feature emerging approaches and technologies to aid further exploration of the genomes from organisms that span the tree of life.

Résumé Biochemical, genetic and physiological mechanisms of aging and age-related changes in humans and animals are presented at the Biology of Aging Gordon Research Conferences. This includes investigations of structure and function that characterize normal aging and investigations of the adverse changes that are risk factors for or accompany age-related disease states. A central objective of the aging conference is to stimulate awareness of the interdependence of genetic, functional and environmental interactions in determining and potentially combating aging. Over the last several years, a rudimentary understanding of some aging mechanisms in cells and simple animals has been generated. In other organisms such as C. elegans, yeast and Drosophila, molecular genetic studies have defined many of the genes that determine life span and gene expression changes associated with aging. In rodents, many biochemical pathways strongly associated aging and life span are now being confirmed and further studied using knockout and transgenic strategies. One of the most exciting advances in our understanding of aging involves the discovery of evolutionarily conserved mechanisms that control longevity, the insulin/insulin-like growth factor pathway in particular. Thus, our field is continuing to mature as we develop a deeper understanding of mechanisms applicable across species. The underlying goal driving the conference is to encourage participants to critically evaluate the latest insights into the aging process from the molecular to the whole organism, with a focus on defining rational approaches to health span extension.

Résumé The existence of the Archaea as the third domain of life was discovered approximately 30 years ago. Since this time, tremendous progress has been made to understand the metabolic pathways and genetic regulons that underlie the often extreme life style of this diverse group of prokaryotes. While certain proteins involved in transcription, translation and replications are shared with the Eukarya, general metabolic diversity resulting in diverse carbon acquisition and energy substrate usage are common with the Bacteria. The Archaea are distinct by exhibiting unique features such as a methane production, unique photosynthesis via a single protein chromophore and the ability to thrive under extreme conditions such as extremely high or low temperature, salt saturation conditions, and extremely high and low pH. Although archaeal organisms represent approximately one third of life's evolutionary diversity and display many interesting and practical properties, Archaea are much less characterized than are organisms from the other two domains. The Gordon Research Conference (GRC) constitutes the most prestigious conference in the field on archaeal research. With the discovery of the methane-producing methanogenic archaea, the first GRC was held on methanogens in 1984 and continued every three years until 1993. At this time, it was recognized that the archaea constitute a diverse group of organisms that were more broadly studied resulting in cutting edge research on novel and unique metabolic features. To reflect the broadening research activities, the first GRC on Archaea was held in 1996 and was named "Archaea: Ecology Metabolism & Molecular Biology". This highly international conference is now held in a two-year cycle and expands on established themes centering on emerging archaea and archaeal viruses and their ecology, extremophilic enzymes and unusual metabolic pathways, mechanisms of archaeal transcription, translation, replication and recombination, and genomics and evolution.

Résumé The Cell Biology of Metals Gordon Conference was initiated to provide a forum for an emerging field, encompassing the uptake, distribution and use of metals in cells and organelles. Metals are taken up across membranes and are directed to various cellular destinations, ultimately finding their way into the correct metalloproteins. Intricate mechanisms control uptake, distribution, trafficking, and insertion into proteins. Transporters, chelators, metal cofactors, protein folding chaperones, metallochaperones, transcription factors and translation regulators are involved. The level of complexity equals that of the protein trafficking pathways that are being characterized by modern methods of cell biology. An important objective is to understand how protein trafficking pathways and metal trafficking pathways are integrated and intersect. Other areas of focus are mechanisms of metal ion homeostasis, how cells sense diverse metal ions, and how homeostatic mechanisms for different metal ions are interconnected. Cellular malfunction or disease ensue if any of the steps in metal homeostasis are perturbed, and the meeting emphasizes these disease related implications. The sessions are organized according to cell structure, cutting across metals (Fe, Cu, Zn, Mo) and evolutionary categories.

Résumé The Zing conference on “Genomic Integrity” will focus on the various pathways that help protect living creatures from the genetic changes that lead to genomic instability, mutagenesis, inherited rare human diseases, cancer, premature aging and neurological disorders. It is now understood that the ability to maintain the integrity of an organism's genome is achieved though the interplay between a variety of DNA repair pathways and the meeting will focus on new insights into our understanding of base excision repair, nucleotide excision repair, mismatch repair, replication fidelity, translesion DNA synthesis, cell cycle signaling and homologous recombination. The program will include time for vibrant group discussions, poster sessions and ample time for individual interactions. The key goal of this international meeting is to allow the community of scientists working in this highly interconnected set of sub-fields to come together in a single location, so as to share their exciting results and to discuss their thoughts for future research directions and collaborations.

Résumé The Angiogenesis Gordon Conference will present the latest research on the basic and translational biology of angiogenesis and its functional role in embryonic/organ development and disease states with specific emphasis on cancer progression/metastasis. Mechanistic concepts and molecular details in vascular biology, blood vessel formation, growth, and regression will be covered. Angiogenesis contributes to the emergence and progression of cancer in a complex manner and unraveling the molecular details will have a significant impact on current and future drug development. We also need to determine the composite consequence of angiogenesis inhibition in the setting of cancer in order to provide optimal information to aid therapy development in the clinic. Angiogenesis research will provide better biomarkers to predict and monitor anti-angiogenesis therapy in our patients. This conference is being designed to address the most vital areas pertinent to angiogenesis, seeking to stimulate interactions among the experts and importantly provide invaluable educational opportunity for graduate students, post-doctoral fellows and junior investigators.

Résumé The study of epigenetics leads to an understanding of genome function. In recent years, epigenetics has revealed some of the most extraordinary forms of gene regulation and quickly became one of the most exciting research areas in genetics and developmental biology. Even as genome projects are providing the complete nucleotide sequence of organism after organism, researchers are turning to the field of epigenetics to understand how that sequence is interpreted in the context of the cell or the organism. Epigenetic modifications such as methylation of the DNA and post-translational modification of core histones affect the organization and packaging of the chromosome. These chromosomal marks are both dynamic and heritable, making epigenetics one of the most intriguing aspects of biology with a wide range of implications for development, disease, and natural variation. These and other aspects of epigenetics research are addressed at the Gordon Research Conference. It has become clear that epigenetic processes encompass an impressive diversity of mechanisms involving noncoding RNAs (ncRNAs), DNA methylation, prions, histone modifications, chromatin remodeling, higher order chromosome structures, and nuclear compartments. The richness and versatility of these biological processes have been elucidated through the study of a wide range of diverse organisms and this conference prides itself on its inclusiveness and its respect for the exchange of ideas and value of learning from the full range of model systems. Apart from advancing our understanding of epigenetic mechanisms by facilitating the exchange of information and ideas from experts working in a wide array of epigenetic systems, the goal of the Conference is to introduce new issues and new questions that push the boundaries of epigenetics and promote the careers of young investigators. Résumé Translational Control has become a major focus of attention in the field of gene regulation and extends to many fields including developmental biology, neurobiology, cell physiology and disease. The discovery of miRNAs opened a new frontier and has been complimented with the finding of new non-coding RNA molecules and the establishment of novel powerful high-throughput techniques, both of which will be central within the conference. With the increasing appreciation for the integration between different steps in the gene expression pathway, the conference will also explore the latest work on the interfaces between translational control and mRNA turnover, RNA localization and splicing/nonsense-mediated decay. The conference will continue to promote communication between experts in the field of translation with those from a scientifically related field that have traditionally held separate meetings on the structure and function of the ribosome. Finally, our biochemical understanding of the translation process is progressing rapidly, and is uncovering hidden layers of regulation including isoforms of translation initiation factors with distinct activities and initiation at non-AUG codons. The latest findings regarding these processes will be featured prominently within the conference. We thus believe that the conference promises to be timely, cutting-edge and of broad interest.

It will cover presentations from cancer genome projects, the areas of cancer functional genomics, systems biology, cancer mouse models, and the translation and clinical impact of scientific results obtained.
