Résumé The analysis of bacterial genomes is revolutionising clinical microbiology, and the clinical microbiologists of the immediate future will have to process and interpret data very different from that provided by agar-centric clinical microbiology laboratories. This course combines practical hands-on laboratory and bioinformatics work with faculty lectures by, and discussions with, leaders in this exciting and expanding area. The principal aim is to equip clinical scientists, specialist registrars, and consultants in medical microbiology (including joint medical microbiology and infectious diseases) with sufficient understanding of these areas to meet these challenges in the future. The course will concentrate on techniques that are currently practicable in diagnosis treatment monitoring, public health and hospital epidemiology and will examine the application of whole genome sequence analysis to clinical microbiology practice.

Résumé Most solid tumors present as dense fibrotic masses, which suggests that fibroblasts contribute to tumor growth by infiltrating and depositing extracellular matrix proteins. Fibroblasts act in wound healing, angiogenesis and tissue remodeling by releasing growth factors and proteases such as matrix metalloproteinases. Therefore, if the growing tumor can co-opt such fibroblasts it has an unlimited source of many of the fundamental elements required for growth and invasion. The two central themes of this workshop are: - Heterogeneity (be it phenotypic, signaling or genotypic), and - Microenvironment (ECM, nutrients, fibroblasts and immune cells). Since a highly heterogeneous tumor has the potential to adapt to any microenvironment, understanding how interactions between the growing tumor and its microenvironment modulate tumor heterogeneity is critical to unraveling the mechanisms of cancer initiation.

Résumé This laboratory-based training course will provide a comprehensive overview and practical laboratory experience of the genetic manipulation of mammalian stem cells.Focusing on mouse ES and human iPS cells, genome informatics, recombineering, CRISPR designer nucleases, transposon technology and gene targeting will be covered in interactive demonstrations, discussions and lectures by international experts. Laboratory work includes the culture and transfection of mouse ES and human iPS cells, design and construction of gene targeting vectors, and the use of PiggyBac transposons and designer nucleases (CRISPRs) for advanced genome engineering and genetic screens. Participants will be trained in the informatics and practical use of public IKMC (International Knockout Mouse Consortium) mouse knockout resources, including the production of mutant mice.Emphasis is placed on the planning/design and successful experimental execution of gene targeting projects and participants should gain the skills necessary to design, construct and target their own genes of interest.

127.EMBO Practical Course — In vivo plant imaging

Résumé This course will teach participants the practical aspects of live-imaging plant tissues. Plants are quite different to animals in many ways. For instance they typically develop much more slowly and cells do not migrate. Their overall morphologies are also very different and the presence of cell walls presents challenges optically. Over many years, certain laboratories have developed specialised imaging techniques optimised for plants. However despite these advances, the adoption of these techniques by the wider plant research community has been extremely limited. These specialised techniques will be taught to help promote their wider adoption. The course will cover the physical preparation of samples from different parts of the plant as well as time-lapse imaging strategies and image analysis approaches and uses.

Résumé Fluorescence is a light-based molecular phenomenon that is inherently non-invasive; it thus finds broad application in the studies of living matter - from the observation of isolated proteins in buffer to complex machinery in living animals. Detection of fluorescence offers highest signal to noise, such that even single molecules can be investigated. This coined the development of myriads of fluorescence-based experimental strategies in biology and biophysics. Progress in biological research is directly linked to the development and subsequent application of new spectroscopic/ microscopic techniques. Researchers in biology are mostly question driven – their aiming at using novel light-microscopy often reflects the hope to gain new insights, based on the promises of publication on the methods. This typically causes a misperception, e.g. that certain experiments performed under controlled biochemical conditions might not be easily adaptable to studies in cells, or that high-resolution methods do frequently not match the demands of in cell studies. Vice versa, in cell studies frequently lack the insights into the molecular mechanism. To achieve the necessary understanding to integrate seamlessly fluorescence tools across different resolution levels and laboratory scales (biochemical-cell-organism) in their research projects, the participants will receive hands on training and specialized training across a large range of high-resolution fluorescence studies. In particular, the course aims to teach the common concepts but also what requirements distinguish a large set of advanced fluorescent spectroscopic and microscopic techniques, from biochemical conditions to complex cellular systems. Experts in different fields will introduce principles, challenges and guidance of their fields. In this surrounding the participants will be able to directly perform small research projects in teams of two people on state of the art equipment and discuss their results with invited investigators, instructors, local teachers and editor.

Résumé The causative links between genomic and genetic variations and cancer are well documented and have provided a greater understanding of tumorigenesis and cancer progression. Molecular genetic information has been used to derive new therapeutic regimens and has accelerated the development and application of "personalised cancer medicine". There are also an increasing appreciation that epigenetic aberrations that are often directly caused by genetic defects resulting in loss- or gain-of-function of epigenetic-regulators also contribute significantly to cancer onset and progression. Indeed, parallel integration of our knowledge of the cancer genome and epigenome using sophisticated "omics-based" technologies and high throughput functional screening techniques are providing even more tangible links between genetic/genomic aberrations, epigenetic/epigenomic dysregulation and tumorigenesis. This information is being functionally validated using experimental models that accurately reflect the underlying genetic and epigenetic alterations observed in human tumor samples. Moreover, the information is being translated into clinical use through the development of genetic and epigenetic biomarkers of cancer development, evolution, heterogeneity and response to therapeutic intervention. This GRC series focuses on the advances in discovering and validating "driver" mutations in cancer genes, understanding the downstream consequences of these genetic changes on the epigenome and how this information is being best utilized to advance cancer detection, monitoring and therapy.

Which signaling and other regulatory networks are altered in tumors relative to the normal tissues, and how do they function within the tumor? Finally, there is growing evidence that therapies aimed at the major cell types in tumors may sometimes make things worse, by leading to an expansion in the fraction of cancer stem cells. How can this be avoided? This workshop will address these and other questions through discussions among mathematical and computational modelers and experimentalists. In particular, the strong connections between normal development, tumor growth and the use of novel treatment strategies will be discussed.

Résumé The Gordon Research Conference (GRC) on the Cell Biology of Megakaryocytes and Platelets is the premier scientific meeting focused on the biology of megakaryocytes and the platelets that arise from them. This will be the 6th biennial meeting that was organized to bring together two groups of scientists with complimentary goals and interests. Fusing new developments in basic science with high-impact translational research, the meeting will provide a stimulating program featuring an international mix of established and upcoming researchers. The meeting will start with two Keynote speakers, Drs. John Hartwig and Shaun Jackson, covering ground-breaking aspects of megakaryocyte and platelet biology. Meeting topics will include emerging roles for platelets in biology, including immunity, tumor metastasis, and lymphatic development. Identification of new signaling pathways that regulate megakaryocyte development and platelet activation will also be a key theme, including an original session focused on membrane biology. Novel mechanisms for growing platelets in vitro as well as innovative ways to visualize and study megakaryocytes and platelets will also come under the spotlight. The conference will also report on the latest findings in the regulation of platelet numbers. In addition, there is an entire session devoted to presentations of Late-Breaking Topics, selected from submitted abstracts. Building on the highly creative, congenial interactions that characterized the previous meetings in this series, this conference will provide the ideal setting for cross-fertilization of ideas, conceptual advances, and the promotion of synergistic collaborations.

Résumé Antimicrobial peptides (AMPs) are essential components of host defense against infections present in all domains of life. These peptides display marked activity against bacteria, fungi, viruses and parasites. But it has become increasingly clear however that AMPs and structurally related peptides also possess functions distinct from this broad-spectrum antimicrobial activity. These include realisation of roles in inflammation, immunity, wound healing and also fertility. The focus of the 2015 Gordon Conference of Antimicrobial Peptides is clear from its subtitle: basic and translational aspects. These aspects will be discussed by invited speakers, but also by scientists that submit abstracts to the meeting. The conference is aimed at scientists from a diverse array of disciplines, including microbiology, biochemistry, animal and plant biology, medicine, immunology, cell biology and drug development. The Conference will address the most innovative and up-to-date science on a range of topics in the field of AMPs research, and seek to integrate the different perspectives on the role of AMPs. AMPs play a major role at epithelial surface, where commensal and pathogenic micro-organisms are present. The intriguing insights provided by analysis of the microbiome at these epithelial surfaces will therefore have a major impact on our understanding of the role of AMPs at these surfaces. Other sessions will be focused on: regulation of expression of AMPs; microbial mechanisms to prevent the cytotoxic activity of AMPs; and their role in health and disease. Application of AMPs as therapeutics, and opportunities for use in agriculture and infection control will also be discussed. The informal and collegial atmosphere of the Conference is an ideal environment for scientific discussions and brainstorming, in formal and informal settings throughout the day. The presence of scientist from different disciplines offers opportunities for discussions that few meetings provide, in and outside the unique "Late-Breaking Topics Corner".

 Résumé     The 2015 Gordon Research Conference (GRC) on "Cannabinoid Functions in the CNS: From Molecules to Disease Mechanisms" will celebrate the 50th anniversary of the discovery of the chemical structure of Δ9-tetrahydrocannabinol, the major psychoactive constituent of Cannabis sativa. This conference will also mark the 25th anniversary of cloning the "neuronal" CB1 cannabinoid receptor. Thus, the meeting will blend the review of long-held conceptual milestones that provided molecular fundaments for endocannabinoid research performed today with disseminating and discussing the newest, groundbreaking results among students, junior faculty and senior investigators. Besides capturing physiological roles for endocannabinoids, ranging from secondary metabolism, synapse physiology to developmental biology including stem cells, this GRC will also serve as a platform for presenting key insights in fear, anxiety, psychosis, addiction, reward, aging and neurodegeneration. Our stated aim is to emphasize molecular mechanisms and comprehensive studies establishing causal relationships to endocannabinoid signaling. Significantly, this topical GRC will feature introductory lectures from world-renown investigators outside the immediate endocannabinoid field to promote interdisciplinarity, interactions, and the discussion of theoretical and technological advances pertinent to advancing endocannabinoid neurobiology. As such, the opening lecture will feature Tamas L. Horvath, while plenaries at the various sessions include Elena Cattaneo, Karl Deisseroth, Andreas Luthi, Raphael Mechoulam and Pier Vincenzo Piazza.  Résumé     Ca2 is a ubiquitous intracellular messenger; it is an essential regulator of many cellular processes including fertilization, cell death, sensory transduction, muscle contraction, motility, exocytosis and fluid secretion. Defective Ca2 signalling is a feature of diverse diseases (e.g. hypertension and immunodeficiencies). Since Ca2 is widely used by cells to regulate their behaviour, there is a correspondingly diverse range of activities amongst investigators with an interest in Ca2 signalling. Thus, this GRC provides a unique opportunity for such investigators, who might otherwise never attend the same conferences, to meet and discuss progress from many perspectives. An important feature of the programme is that it brings together investigators whose primary interests are in fundamental aspects of Ca2 signalling with those whose interests focus primarily on the physiology or pathology of specific tissues in which Ca2 signals play an important role. The selection of speakers and topics for the sessions reflects this diversity. For example the topics that are addressed in this conference include plasma membrane signal transduction/sensing mechanisms, intracellular calcium release, calcium entry, calcium channel regulation and structure, calcium pump function, signalling microdomains, and disease associated with calcium signalling.  Résumé     Unlike many excellent specialty meetings where experts in eukaryotic or prokaryotic DNA replication, recombination, or transcription delve in to the intricacies of their special realm, the Chromosome Dynamics GRC takes a different tack. It includes interested participants with very different specialties and methodologies who share the common goal of understanding chromosome dynamic and the mechanisms that organize large DNA molecules. This is critical because homologous and site-specific recombination, transposition target selection, adaptive mutation, gene transcription/silencing, gene order on chromosomes, and genome segregation are all phenomena that depend on accurate models of chromosome dynamics. Breakout work on chromosomes from viruses to man has been announced at past meetings. Highlights included: new biochemistry and cell biology of condensins and cohesins, new roles for telomeres in replication and repair, single-molecule studies of chromatin remodeling proteins, mechanisms of triplet repeat expansions in diseases like Huntington’s Chorea and fragile X syndrome, and new roles for RNA in controlling epigenetic gene regulation and centromere function.

 Résumé     In the past decade it has become clear that regulatory RNAs allow bacteria and archaea to response to a wide range of changes in their environmental conditions, and that much more of the genome is transcribed than ever imagined. This conference is the fourth in a series of extremely successful meetings exploring the latest findings regarding this important class of regulators. The conference will be a premier forum for the presentation of cutting-edge advances and the latest perspectives in the areas of discovery, mechanism, structure and evolution of bacterial riboregulators. Sessions will cover prominent and emerging roles of small RNAs in regulatory networks as well as their diverse mechanisms of action. Other sessions will address how RNA-binding proteins and ribonucleases impact sRNA function. Riboswitches and other signals within mRNA that affect the stability or translation of the message as well as the important RNA-based CRISPR immunity against invading genetic elements also will be discussed. Finally, regulatory RNAs will be considered from broader evolutionary, systems and synthetic biology perspectives. In addition to featuring scientists invited from around the world, the program will include 20 speakers selected on the basis of submitted abstracts. This flexibility in programming allows inclusion of exciting late-breaking advances and will promote high visibility of rising young investigators. This meeting aims at bringing together researchers of all stages using diverse approaches to study many aspects of RNA regulation in a variety of systems, the main goal being to share novel insights, establish collaborations and facilitate cross-fertilization of ideas among investigators, postdoctoral fellows and graduate students.

The cell cycle involves a complex series of events, including cell growth, genome duplication, and mitosis. Dramatic progress in these areas has come from experiments performed in organisms ranging from yeast to mammals using approaches spanning high resolution structure determination to systems-level analysis. To continue this interdisciplinary tradition, an elite group of international researchers using diverse experimental approaches and systems will discuss their latest findings at this meeting. Sessions will focus on basic events underlying cell division. Topics will include: how cells regulate their size and metabolism; how they transit through G1; the molecular machines that carry out DNA replication and how they contend with chromatin structure; how cells sense, signal, and repair DNA damage inside and outside of S phase; how cells prepare for and execute mitosis; and the mechanical events underlying chromosome segregation and cytokinesis.  Résumé     The existence of the Archaea as the third domain of life was discovered approximately 30 years ago. Since this time, tremendous progress has been made to understand the metabolic pathways and genetic regulons that underlie the often extreme life style of this diverse group of prokaryotes. While certain proteins involved in transcription, translation and replications are shared with the Eukarya, general metabolic diversity resulting in diverse carbon acquisition and energy substrate usage are common with the Bacteria. The Archaea are distinct by exhibiting unique features such as a methane production, unique photosynthesis via a single protein chromophore and the ability to thrive under extreme conditions such as extremely high or low temperature, salt saturation conditions, and extremely high and low pH. Although archaeal organisms represent approximately one third of life's evolutionary diversity and display many interesting and practical properties, Archaea are much less characterized than are organisms from the other two domains. The Gordon Research Conference (GRC) constitutes the most prestigious conference in the field on archaeal research. With the discovery of the methane-producing methanogenic archaea, the first GRC was held on methanogens in 1984 and continued every three years until 1993. At this time, it was recognized that the archaea constitute a diverse group of organisms that were more broadly studied resulting in cutting edge research on novel and unique metabolic features. To reflect the broadening research activities, the first GRC on Archaea was held in 1996 and was named "Archaea: Ecology Metabolism & Molecular Biology". This highly international conference is now held in a two-year cycle and expands on established themes centering on emerging archaea and archaeal viruses and their ecology, extremophilic enzymes and unusual metabolic pathways, mechanisms of archaeal transcription, translation, replication and recombination, and genomics and evolution.

Metals are taken up across membranes and are directed to various cellular destinations, ultimately finding their way into the correct metalloproteins. Intricate mechanisms control uptake, distribution, trafficking, and insertion into proteins. Transporters, chelators, metal cofactors, protein folding chaperones, metallochaperones, transcription factors and translation regulators are involved. The level of complexity equals that of the protein trafficking pathways that are being characterized by modern methods of cell biology. An important objective is to understand how protein trafficking pathways and metal trafficking pathways are integrated and intersect. Other areas of focus are mechanisms of metal ion homeostasis, how cells sense diverse metal ions, and how homeostatic mechanisms for different metal ions are interconnected. Cellular malfunction or disease ensue if any of the steps in metal homeostasis are perturbed, and the meeting emphasizes these disease related implications. The sessions are organized according to cell structure, cutting across metals (Fe, Cu, Zn, Mo) and evolutionary categories.

Topics will include advances in molecular dynamics simulation of enzymes; computational approaches to studying the interaction of drugs, drug transporters and signaling enzymes with lipid membranes; simulation of protein-protein interaction and multi-protein complex enzymatic systems; computational study of signaling pathways; new techniques in computational drug discovery as they apply to enzyme reactivity and dynamics; simulation of allosteric interactions; advances in multi-scale modeling; new techniques for direct simulation of reactivity in biological systems; and the impact of technological advances in computer hardware and software on the field of chemical biology. The conference will feature lectures by established leaders in the fields, as well as encouraging young and talented researchers to present both challenging questions and new ideas for solving them. The aim is to encourage a collegial atmosphere to promote stimulating and insightful discussion.

 Résumé     The 2015 GRC on Catecholamines will focus on cutting-edge developments in catecholamine research. In recent years, catecholamines have been the focus of investigations using a broad range of approaches from molecular biology to behavior and clinical studies. One of the characteristics of the field is that neuronal systems using catecholamines have much in common, although investigators who focus on particular catecholamines or on different actions of the same catecholamine are often unaware of complementary aspects of catecholamine research. It is increasingly apparent that continued progress will require increasingly integrated approaches in studies of catecholamine biology, function and dysfunction. Thus, molecular biologists are being drawn to more integrated systems approaches and behavioral biologists are exploiting many molecular approaches. In addition, because DA, NE and epinephrine serve as neurotransmitters and hormones throughout the phylogenetic scale, studies are possible in simple organisms where powerful genetic tools are available. We plan to highlight these new advances by including some of the researchers at the forefront of this field (and many of them can be considered junior scientists) in an attempt to provide a unique opportunity to discuss recent advances in the understanding of how catecholamine systems may contribute to the pathophysiology and treatment of those conditions.

 Résumé     The Gordon Research Conference on Cellular and Molecular Mechanisms of Toxicity is a premier forum for showcasing the latest, most innovative advances in mechanistic toxicological research. For the 2015 conference, the organizers have assembled a group of world-leading experts working in areas of investigation that are highly relevant to environmental, industrial, and pharmaceutical toxicology, with the goal of incorporating research on the mechanistic basis of disease into risk decision making. Emerging technologies in mechanistic toxicology and computational, 3D, and in vivo modeling as well as systems biology approaches to assessing risk to human disease will be highlighted. The microbiome, the inflammasome, epigenetics, carcinogenesis, and stem cells in toxicological research are among the topics that will be highlighted at the conference. The conference will attract a mix of junior and senior investigators and trainees with a common interest in advancing their respective fields of investigation. For the second time, a Gordon Research Seminar on Cellular and Molecular Mechanisms of Toxicity will precede the conference. This provides an expanded opportunity for trainees and young scientists to be exposed to cutting-edge science and to network with their future colleagues in the field. The poster sessions are an essential element to the success of conference. Trainees and scientists at any level will have the opportunity to present their latest, groundbreaking work in poster format under an informal atmosphere that fosters relaxed dialog, networking and idea sharing. A special committee will select some abstracts/posters for short oral talks. These "late-breaking" oral sessions provide increased participation to trainees and young scientists to share their work with an audience of experts. This is a tremendous opportunity for the young talent to receive feedback from well-established leaders in the field. Another key to the success of the Gordon Conference is the social interaction of participants at meal times and during social activities in afternoon breaks. These interactions have led to long-lasting scientific collaborations and the lining up of trainees with laboratories for further education.

 Résumé     Translational Control has become a major focus of attention in the field of gene regulation and extends to many fields including developmental biology, neurobiology, cell physiology and disease. The discovery of miRNAs opened a new frontier and has been complimented with the finding of new non-coding RNA molecules and the establishment of novel powerful high-throughput techniques, both of which will be central within the conference. With the increasing appreciation for the integration between different steps in the gene expression pathway, the conference will also explore the latest work on the interfaces between translational control and mRNA turnover, RNA localization and splicing/nonsense-mediated decay. The conference will continue to promote communication between experts in the field of translation with those from a scientifically related field that have traditionally held separate meetings on the structure and function of the ribosome. Finally, our biochemical understanding of the translation process is progressing rapidly, and is uncovering hidden layers of regulation including isoforms of translation initiation factors with distinct activities and initiation at non-AUG codons. The latest findings regarding these processes will be featured prominently within the conference. We thus believe that the conference promises to be timely, cutting-edge and of broad interest.

Since computational prediction methods continuously improve and evolve towards prediction of structure of more and more complicated biological objects our conference will cover also related topics, such prediction of structure of protein complexes, protein docking; prediction of intrinsically disordered regions in proteins, RNA structure prediction and modeling of RNA-protein assemblages. New methodologies for protein structure and function prediction will be discussed, such as physics-based approaches to the structure prediction, structural flexibility models, and development of new improved coarse-grained potentials.
